You do need to set it after getting the engine up to temperature on the circuit; it will totally change from cold to hot. For a new or rebuilt carb I set the tickover to about 6000rpm when cold, then adjust it after the run. I found using my normal DD mixture settings it was impossible to get it to idle just using the tickover screw, and had to adjust the mixture (TalKo do recommend this, both in the printed guide and when I spoke to them about this problem).
