The reason some slick tyres are directional is due to the way they are made and how the laminates are bonded together. The overlap of the bonded parts determines the direction of tyre. However modern manufacturing processes are so accurate and repeatable that it is not really an issue anymore.
I have run directional slicks both ways and there is no difference that we can see in the times and handeling.
