As has been said it could be the reed block and of course the spark plug.
The problem I have is that if runs OK on the dyno then the only difference is that you are not driving it on the dyno and therefore it could well be you and the way you are trying to drive it.

Also change the fuel and the pulse-pipe back to the standard versions, also check the pipe inside your fuel tank is flexible and not leaking, also while you are at it re-kit your fuel pump.
You said in your first post “It will not idle, even with the idle screw fully in” Now the idle screw is an air valve so if you close it up then you are running too rich, the more you screw it out the leaner you go. The other screw lifts the throttle slide and therefore controls the idle speed.
